Characterization of the PRDM9 Zinc-Finger array binding and multimer formation

PRDM9 has been identified as a meiosis specific protein that plays a major role in determining the location of meiotic recombination hotspots, but so far the binding characteristics of PRDM9 have still been enigmatic. We characterized the binding specificity of murine PRDM9Cst to a target recombination hotspot, as well as the multimer formation, using EMSAs. Moreover we tested different PRDM9 constructs missing distinct domains to also assess what regions of PRDM9 induce multimerization. In these experiments we demonstrated that PRDM9 forms functional multimeric complexes of at least two or more monomer units that are mediated within the C-terminal Zinc-Finger domain.
