Characterization of extrinsic protein PsbP from Photosystem II by solution NMR spectroscopy

Psb proteins of photosystem II (PSII) are a class of extrinsic proteins located in the cytosolic part of the oxygen evolving complex (OEC). They help to maintain proper conditions for the water splitting reaction ultimately leading to the release of molecular oxygen1. Each of these proteins (PsbP, PsbQ, PsbO) contains function- ally indispensable intrinsically disordered regions (IDRs), which are not resolved in X-ray diffraction data. Solution NMR spectroscopy therefore is the preferred ap- proach to assess their structural propensities, dynamics and interactions. We have determined three-dimensional structures of PsbP (23 kDa) and PsbQ (16 kDa) in solution including their highly flexible IDRs, which were incompletely characterized by other structural biology techniques (X-ray, cryo-electron microscopy)2-6. The dynamic interactions of the Psb proteins have been studied using combination of NMR techniques such as chemical shift perturbation (CSP) titration and chemical exchange saturation transfer (CEST) as well other methods like isothermal titration calorimetry (ITC) and microscale thermophoresis (MST).
